Incorporated 1867, the Indianapolis, Cincinnati, and Lafayette Railroad eventually went bankrupt and merged with other railroad companies as noted below.

The Indianapolis, Cincinnati, and Lafayette Railroad went bankrupt in 1880 and was reorganized as the Cincinnati, Indianapolis, St. Louis, and Chicago Railway (CISL&C). The CISL&C operated a railroad line from Cincinnati via Indianapolis to Lafayette, being the result of an 1867 merger of the Indianapolis and Cincinnati Railroad (I&C), the Lafayette and Indianapolis Railroad (L&I), and the Cincinnati and Indiana Railroad (C&I). The three predecessor companies had been founded in 1850, 1846, and 1861, respectively.

The railway line, originally between Indianapolis and Lafayette, was built by the Lafayette and Indianapolis Railroad in 1850. Over the years and due to mergers, the line was owned by several railroads during its operation:
- Indianapolis, Cincinnati, and Lafayette Railroad, 1867
- Cincinnati, Chicago, and St Louis Railroad, 1880
- Cleveland, Cincinnati, Chicago, and St Louis Railroad (the Big Four), 1889
- New York Central Railroad, 1930
- Penn Central Railroad, 1968
- Conrail, 1976

The route's modern-day equivalent is the CSX and Norfolk Southern lines that operate freight service today.
